preparation | add 7-azaindole to 10ml dichloromethane, slowly add glacial acetic acid Dropwise at 0 °c, stir for 20min. Then, 5.6g(37mmol) of phosphorus oxychloride was dissolved in 20ml of dichloromethane, and the mixture was added dropwise to the reaction system for 2H at 0 °c. The progress of the reaction was monitored by thin layer chromatography, and the reaction was stopped until no reactant was found. TLC detection (methanol developing solvent) showed that the product was Rf = 0.57. After completion of the reaction, a saturated aqueous solution of sodium sulfite was added to the reaction system until the starch KI test paper did not turn blue. Dichloromethane was concentrated, 20ml of water was added, and NaOH was added to adjust pH>13 at 0 °c. The reaction mixture was solvent-dried, dissolved in dichloromethane, and washed twice with purified water, the organic phase was dried over anhydrous sodium sulfate, the mixture was subjected to silica gel column chromatography, and the product was dried in vacuo to give 4-chloro-1h-pyrrolo [2,3-B] pyridine-2 (3H)-one. |
